This book, like other monographs of the Cellular Organelles series, is not a comprehensive review, but an introduction to the study of cytoskeleton. Accordingly, we describe only the main facts and concepts related to cyto skeleton. Needless to say, selection and interpretation was influenced by the personal interests and opinions of the authors, although we attempted to be as fair as possible. We wished to familiarize the reader not only with well established facts, but with current unsolved problems. Therefore, the words "possibly," "maybe," "not known," and "not clear" are much more frequent in this text than in many others. In accordance with the style of the series, relatively short lists of addi tional readings are given at the end of each chapter; these lists contain mostly the recent reviews and a few original papers describing certain phenomena in detail. Few references are cited in the text; these citations are given to help the reader find the source of certain new data and theories, which are not discussed at length in the reviews. In contrast, many well-established facts and widely known theories are not cited.

Our current under standing of the three-dimensional organization of the cytoplasm involve three major fibrous proteins which are collectively known as the cytoskeletal system. These polymorphic cytoskeletal proteins are microtubules (25-nm diameter), microfilaments (6-nm diameter), and intermediate filaments (lO-nm diame ter). Microtubules consist of tubulin and several well-characterized micro tubule associated proteins (MAPs) including MAP , MAP , tau, and others. l 2 Microfilaments consist of actin and associate with actin-binding proteins in cluding a-actinin, filamin, myosin, tropomyosin, vinculin, and others. Inter mediate filaments (lO-nm filaments) consist of at least five different tissue specific classes, including desmin or skeletin (muscle), prekeratin (epithelial), vimentin (mesenchymal), neurofilament (nerve), and glial acidic fibrillary protein (astrocytes). These major fibrous proteins apparently interact with each other as well as other cytoplasmic components and appear to be inti mately associated with such biological processes as cell shape changes, growth, motility, secretion, cell division, and uptake of materials from the exterior of the cell.

The conference provided an in-depth discussion of the effects a~d mechanism of action of some toxic agents on the cytoskeleton. Mamma"lian and other eukaryotic cells contain protein networks within the cytoplasm comprised of microfilaments, intermediate Hlaments and microtubules. These components of the cytoskeleton playa key role in cell shape, motility, intracellular organization and transport, and cell division. Furthermore, the cytoskeleton, via associations with the cell membrane, appears to function in intracellular communication and cellular responses to membrane events. Because of the complex functional roles of the cytoskeleton which vary with cell type, degree of differentiation, and cell cycle, its disruption may result in a variety of cellular changes. This expanding field in cell biology has already attracted the interest of toxicologists and environmental health scientists as a potentially fruitful area of research. Indeed, there is mounting evidence that certain toxic and chemotherapeutic compounds, as well as physical agents such as radiation and hydrostatic pressure, disrupt the normal structure and function of the cytoskeleton. This may be an important step in the overall expression of their action. It was, therefore, an opportune time to hold a conference to encourage the development of this area of toxicology and to suggest directions for future research.

Cross-commu- cation among cytoskeletal polymers is thought to be achieved through cytoskeletal polymer accessory proteins and molecular motors that bind two or more cytoskeletal polymers. Development of the modern concept of the cytoskeleton is a direct o- growth of advances in experimental tools and reagents that are available to cell and molecular biologists. Technological advances and refinements in cell imaging have made it possible to selectively image a single cytoskeletal po- mer and monitor its dynamics through the use of fluorescence probes in vitro and in vivo. Two decades ago, cytoskeletal research was limited to a few perturbation reagents that included colchicine and cytochalasin. Today, the perturbation arsenal has expanded to a highly selective group of reagents that includes Taxol, nocodazole, benomyl, latrunculin, jasplakinolide, and such endogenous proteins as gelsolin. These reagents enable the investigator to selectively perturb or destroy a cytoskeletal polymer while leaving other cytoskeletal polymers intact. Site-specific monoclonal antibodies that target a specific cytoskeletal polymer have proven to be highly selective affinity tools for cytoskeletal research.
